Identification and characterization of a sulfite reductase gene and new insights regarding the sulfur-containing amino acid metabolism in the basidiomycetous yeast Cryptococcus neoformans

(Submitter supplied) The amino acid biosynthetic pathway of invasive pathogenic fungi has been studied as a potential antifungal drug target. Studies of the disruption of genes involved in amino acid biosynthesis have demonstrated the importance of this pathway in the virulence of Cryptococcus neoformans. Here, we identified the MET5 (CNL05500) and MET10 (CNG03990) genes in this pathway, both encoding sulfite reductase, which catalyzes the reduction of sulfite to sulfide. more...

Transcriptome of Cryptococcus neoformans and Cryptococcus deneoformans

(Submitter supplied) The goal of this study was to position all transcripts extremities in two species of Cryptococcus using TSS-Seq and QuantSeq 3' mRNA-Seq when cells are grown under different conditions. We analysed also the level of expression of each genes in the same condition using the same cell sample. All these data have spiked in using a fixed quantity of S. cereviae cells added just before DNA and RNA extraction.

Cryptococcus neoformans sexual reproduction is controlled by a quorum sensing peptide

(Submitter supplied) Mating pheromone is the best-known fungal paracrine signal whose essentiality in determining sexual reproduction has been recognized since six decades ago. We demonstrate that the pheromone cannot induce mating response during unisexual mating due to the lack of a compatible receptor from the same unisexual population. We identify a quorum sensing (QS) peptide Qsp1 in place of pheromone as a specific intercellular signal directing Cryptococcus unisexual reproduction. more...

Time-course microarray study of Cryptococcus neoformans var. neoformans spore germination

(Submitter supplied) To identify the genome-wide transcriptional changes that occur throughout germination of C. neoformans spores, we conducted a time-course microarray experiment spanning six timepoints to generate a temporal expression pattern for each known gene. Spores were placed in rich medium and allowed to germinate for 10 hours, when they start to replicate as yeast. Each time point was flash frozen in liquid nitrogen and RNA was harvested for each time point at the same time. more...

Microarray study of the Sex Inducer Proteins in Cryptococcus neoformans var. neoformans (JEC20/21)

(Submitter supplied) To identify genes regulated by the homeodomain transcription factors Sxi1α and Sxi2a (Sex Inducer 1α and 2a) in Cryptococcus neoformans, we carried out a whole-genome expression experiment. We performed two independent whole-genome microarray experiments comparing transcript levels in cells that either possessed or were lacking the Sxi proteins. In the first experiment we compared the expression profile of a haploid sxi1αΔ strain to that of a haploid strain expressing both SXI1α and an inducible copy of SXI2a (Sxi +). more...

Comparison of transcription profiles of C. neoformans during rabbit or human infection

(Submitter supplied) Ubiquitination is a reversible protein modification involved in various cellular processes in eukaryotic cells. Deubiquitinating enzymes, the proteins responsible for the removal of ubiquitin, act as essential regulators to maintain ubiquitin homeostasis and to exquisitely regulate protein degradation via the ubiquitination pathway. Cryptococcus neoformans is an important basidiomycete pathogen that causes life-threatening meningoencephalitis primarily within the immunocompromised population. more...

WT vs rim101 mutant gene expression under capsule inducing conditions

(Submitter supplied) Cryptococcus neoformans is a prevalent human fungal pathogen that must survive within various tissues in order to establish a human infection. We have identified the C. neoformans Rim101 transcription factor, a highly conserved pH-response regulator in many fungal species. The rim101- mutant strain displays growth defects similar to other fungal species in the presence of alkaline pH, increased salt concentrations, and iron limitation. more...

Light-regulated genes in Cryptococcus neoformans

Effect of Nrg1 on pathogenesis of Cryptococcus neoformans

(Submitter supplied) The Cryptococcus neoformans NRG1 gene was identified using gene microarrays to define putative transcription factor genes regulated by the cyclic AMP (cAMP) signal transduction pathway. Disruption of NRG1 results in delayed capsule formation and mating, two phenotypes that are directly controlled by cAMP signaling. Putative targets of the Nrg1 transcription factor were identified using a second genome microarray to define differences in the transcriptomes of the wild-type and nrg1 mutant strains. more...

rdi1∆ vs WT during macrophage co-culture

(Submitter supplied) Rho-GDP dissociation inhibitors (RDI) are repressors of Rho-type monomeric GTPases that allow for precise control of their target processes, e.g. cytoskeletal arrangement, vesicle trafficking, and polarized growth. In the human pathogenic yeast Cryptococcus neoformans, maintenance of normal cell morphology is vital for pathogenicity. We identified and deleted the gene encoding an RDI homolog in the human fungal pathogen Cryptococcus neoformans and investigated its impact on pathogenicity in animal models of cryptococcosis. more...
