Regulation of the circadian clock by the metabolic enzyme AHCY

(Submitter supplied) Mammalian circadian rhythms are based on coupled transcriptional-translational feedback loops driven by the transcription factors CLOCK and BMAL1. Chromatin remodeling mechanisms are essential for the proper timing and extent of circadian gene expression. We report that the S-adenosylhomocysteine (SAH) hydrolysing enzyme AHCY binds to CLOCK-BMAL1 at chromatin and drives circadian transcription by promoting cyclic H3K4 trimethylation and recruitment of BMAL1 to chromatin.

Determinants of sensitivity to DZNep induced apoptosis in multiple myeloma cells

(Submitter supplied) The 3-Deazaneplanocin A (DZNep), one of S-adenosylhomocysteine (AdoHcy) hydrolase inhibitors, has shown antitumor activities in a broad range of solid tumors and acute myeloid leukemia. Here, we examined its effects on multiple myeloma (MM) cells and found that, at 500 nM, it potently inhibited growth and induced apoptosis in 2 of 8 MM cell lines. 3-Deazaneplanocin A effect on multiple myeloma cells

Analysis of MM cell lines (KMS18, MM.1S, NCI-H929, OPM-2) exposed to 500nM 3-Deazaneplanocin A (DZNep). DZNep potently inhibited growth and induced apoptosis in the NCI-H929 and MM.1S cell lines. Results provide insight into the molecular mechanisms underlying DZNep-induced apoptosis in MM cells.
