Gene expression in human keratinocytes from psoriatic skin (directly into the lesion or in area without lesion) or healthy person (control) used for human tissue-engineered skin substitutes and co-cultured with T-lymphocyte

(Submitter supplied) The objective of this study was to find deregulated genes between healthy and psoriatic T cell-enriched tissue-engineered models to develop a new therapeutic pathway in psoriasis treatment. In this study, we used a tissue-engineered, two-layers (dermis and epidermis) human skin substitute enriched in T cells as a biomaterial to study both the cellular and molecular mechanisms involved in psoriasis’ pathogenesis.

Effect of the primed exposure of Diesel CAST model aerosol in air-liquid interface BEAS-2B cells exposed to native birch pollen

(Submitter supplied) BEAS-2B cells, at air-liquid interface, were exposed to Diesel CAST model aerosol in an vitro exposure system and, later, to native birch pollen using a Pollen Sedimentation Chamber stabilished before. The same exposure was performed, without the primed anthropogenic exposure. The goal was to understand the effect of pre-exposure to a model diesel aerosol in allergic sensitization, using a model stabilished that mimics a real life exposure as closer as possible.

Novel Peripheral Blood Allergen Stimulation Test for Non-IgE-mediated Gastrointestinal Food Allergies in Neonates and Infants.

(Submitter supplied) The lymphocyte stimulation test (LST) helps with the diagnosis of non-IgE-mediated gastrointestinal food allergies. However, The LST requires large volumes of fresh peripheral blood and long culture times. The purpose of this study was to develop a novel LST. we obtained whole blood from patients with non-IgE-GI-FAs to cow’s milk and disease control children. And, we stimulated it with α-casein and Pmix, which is a mixture of four milk protein components (α-casein, κ-casein, α-lactalbumin, or β-lactoglobulin for 24 hours. more...

Gene expression in human corneal epithelial cells (hCECs) with different post-mortem intervals and cultivated in monoculture or used for human tissue‐engineered corneas (hTECs)

(Submitter supplied) In this study, we evaluated the effects of the post-mortem interval, defined as the number of days elapsed between the death of a donor and the isolation of hCECs, on the properties of hCECs. Using gene profiling, we compared the mRNA profiles of passage 1 hCECs (10 populations) cultivated in moculture with different post-mortem intervals and passage 2 hCECs (6 populations) with different post-mortem intervals from hTECs.

Gene expression in human keratinocytes cultivated with serum-free medium and serum containing medium

(Submitter supplied) The objective of this study was thus to develop ab initio that chemically defined serum-free medium. We used a statistical design of experiment (DOE) approach to screen a large number of candidate factors, for both their potential individual and synergetic effects, as to identify a formulation which was then tested extensively. In this study, we present the properties of keratinocyte cultivated with serum-Free medium (Surge SFM) and complemented DME-Ham medium (with serum).

PCSK9 is involved in human induced pluripotent stem cell proliferation through the regulation of the NODAL signaling pathway II

(Submitter supplied) Proprotein convertase subtilisin kexin type 9 (PCSK9) is a key regulator of LDL cholesterol metabolism and the target of lipid-lowering drugs. PCSK9 is mainly expressed in hepatocyte. Here, we show that PCSK9 is highly expressed in undifferentiated hiPSCs. PCSK9 inhibition in hiPSCs with the use of shRNA, CRISPR/cas9-mediated knockout or endogenous PCSK9 loss-of-function mutation R104C/V114A unveiled its new role as a potential cell cycle regulator through the NODAL signaling pathway. more...

CD271 is preferentially expressed and essential for cell proliferation in squamous cell carcinoma of the lung

(Submitter supplied) Squamous cell carcinoma is a major type of cancer in the lung. While several therapeutic target molecules for lung adenocarcinoma have been identified, little is known about lung squamous cell carcinoma (LSCC). We recently reported that CD271 (p75 neurotrophin receptor) serves as a marker for tumor initiation and is a key regulator of cell proliferation in hypopharyngeal squamous cell carcinoma. In this study, we found that CD271 was also expressed in squamous cell carcinoma, but not in adenocarcinoma, of several tissues, including the lung. more...

microarray Analysis of MKN-45 cell lines after 48 hours post-transfection of miR-125b mimic (50nM) and mimic control.

(Submitter supplied) We demonstrate that miR-125b, a key node in this microRNA regulatory network, is upregulated in gastric cancer (GC) and associated with poor overall survival through an integrated analysis of microRNA and mRNA profiling of GC revealed a mRNA-regulatory network. So we have employed whole genome microarray expression profiling as a discovery platform to compare the transcriptome profiling of human gastric cells (MKN-45) after 48 hours post-transfection of miR-125b mimic (50nM) and mimic control.Pathway analysis shows that the predicted targets of miR-125b are highly involved in apoptosis/program death pathway, and the robust apoptosis genes, BIK and CASP6 are validated as the directed targets of miR-125b.

Allergen-specific immunotherapy induces the suppressive secretoglobin 1A1 in cells of the lower airways

(Submitter supplied) While several systemic immunomodulatory effects of allergen-specific immunotherapy (AIT) have been discovered, local anti-inflammatory mechanisms in the respiratory tract are largely unknown. We sought to elucidate local and epithelial mechanisms underlying allergen-specific immunotherapy in a genome-wide approach. We induced sputum in hay fever patients and healthy controls during the pollen peak season and stratified patients by effective allergen-immunotherapy or as untreated. more...
