GPCR-induced activation of PLC-γ by PTKs. An agonist-occupied GPCR induces the exchange of GDP for GTP on the α subunit of Gs or Gi and the subsequent dissociation of the α subunit from the membrane-associated Gβγ dimer. The GTP-bound αs or αi subunit activates c-Src by inducing its autophosphorylation, and the phosphorylated enzyme then associates with and phosphorylates a receptor PTK. The phosphorylated receptor PTK provides a structural scaffold for the assembly of a signaling complex that includes PLC-γ (see Figure 4).
