Each well contains DNA from an entire 96-well plate and is at a complexity of 4000 genomes. Use these pooled DNA preps as DNA templates for the primary library screen.

The promoter::gfp vector is generated by a multi-site LR reaction between pENTRY-prom, pENTRY-gfp and pDEST-DD03. attL sites recombine with corresponding attR sites. The resulting plasmid, promoter::gfp, contains the promoter and GFP flanked by attB sites.
