Adenovirus mediated, heat-regulated gene therapy in a mouse melanoma model. The HSP70 promoter was used to drive gene expression of IL-12. Experimental tumors were established in syngeneic C57BL6 black mice by implanting 10⁶ tissue cultured B16F10 melanoma cells. Viral injections were carried out 1 week later when tumors grew to sizes of 5 to 7 mm in diameter. Mice were injected intratumorally with adenoviruses encoding (1) a heat inducible EGFP gene alone (_•); (2) a heat-inducible EGFP gene with heat treatment (□); (3) the murine IL-12 gene (▴) alone; and (4) the murine IL-12 gene with heat treatment (Δ).
