Model showing the regulation of RetGCs by GCAPs. Particulate RetGCs are functional as dimers. GCAPs are thought to constitutively bind to RetGCs, and regulate cyclase activity by structural changes caused by the binding or dissociation of Ca²⁺. The Ca²⁺loaded form of GCAPs (predominant in resting photoreceptors in the dark) would stabilize the inactive conformation. Dissociation of Ca²⁺ from the GCAPs in response to light would promote dimerization of the cyclase catalytic domains, and cGMP synthesis. ECD, extracellular domain; KHD, kinase homology domain; CAT, catalytic domain.
