This sequence is derived from a suppressive subtraction cDNA library enriched in genes from temperate Oxytropis species. cDNA was synthesized (using SuperSmart PCR cDNA Synthesis kit, Clontech) from mRNA extracted from temperate Oxytropis campestris and O. splendens plantlets grown on petri plates in temperate controlled conditions. Subtraction was carried out using the PCR-select cDNA subtraction kit (Clontech) and cDNAs from arctic Oxytropis arctobia and O. maydelliana plantlets grown on petri plates in arctic controlled conditions. The remaining cDNAs were non-directionally inserted into pCR2.1TOPO vector (Invitrogen) and used to transform ElectroMAX DH10B competent cells (Invitrogen).
