Stratagene (cat. #200450); Total RNA was extracted separately from leaf tissues, using the method described by Chung et al. (2009). mRNA was reverse transcribed into double stranded cDNA using a modified oligo (dT)18 primer with an identifying tag sequence. Double stranded cDNAs were size selected (more then 500 bp), adaptored with EcoRI adapters at both ends and then digested with XhoI. The cDNAs were then directionally cloned into EcoRI-XhoI digested pBSII SK(+) phagemid vector (Stratagene). The total number of white colony forming units (cfu) was 0.8x100,000,000 cfu. The background of empty clones was less than 10%. Inserts ranged from 0.3 kb to 5kb, as determined by PCR.
