cDNA library was derived from vegetative and reproductive shoot tips of Agapanthus praecox ssp. orientalis. cDNA synthesis was initiated using a oligo(dT) primer. Double-stranded cDNA was blunted, digested with EcoRI and MseI restriction endonuclease, ligated to EcoRI and MseI adaptors, using cDNA-AFLP to screen the different expression gene, and cloned into the pMD18-T simple vector.
