RNA was isolated from plants 48 and 72 h post infection with Colletotrichum graminicola. After purification of mRNA first strand cDNA synthesis was performed, and infection-specific cDNAs were obtained after subtractive hybridization with photobiotinylated mRNA from vegetative hyphae and healthy maize leaves. The infection-specific cDNAs were subjected to second strand cDNA synthesis, and these cDNAs, representing putative differentially ESTs, were cloned into pCR 4-TOPO.
