To further explore the mechanism of how MIANGUA2 reduce lung inflammation, we collected the lung tissues to conducted the transcriptome analysis at day 7 post infection. The GSEA analysis showed that the downregulation of TNF, Apoptosis and NF-Kappa B signaling pathway in the MIANGUAN2 group compared with Virus group. In the study, treatment with MIANGUAN2 reduced the expression of Tnfrsf1a and Tnfrsf1b genes, which is consistent with the result that MIANGUAN2 actively down-regulated the TNF signaling pathway . In the Apoptosis pathway, we observed downregulation expression of the apoptotic gene families such as caspases (Casp3, Casp7, Casp8, Casp12) and B cell lymphoma family of genes (Bak1, Bax, Bid) in MIANGUAN2 group compared with Model group. Besides, oral administration with MIANGUAN2 also dampened NF-Kappa B signaling pathway though decreasing the expression of key genes Traf2, Traf3 and RelB. Based on these results, it was concluded that oral MIANGUAN2 can alleviate lung inflammation probably though inhibiting the expression of multiple inflammatory signaling pathways in the lung.
