More...Extended pluripotent stem cells (EPSCs) are stem cells that derive from pre-implatation embryos in specific culture conditions. They can contribute to both embryonic (Em) and extraembryonic (ExEm) lineages. EPSCs derived from mouse and human embryos have been successfully established in the LCDM condition. However, the same has not yet been achieved with porcine embryos. To investigate the species-specific molecular mechanism of porcine embryo derived stem cells (pLCDM) in LCDM condition, we compared the biological characteristics of pLCDM and porcine extpanded potency stem cells (pEPSCs). The results revealed that pLCDM exhibited specific pluripotent traits and limited differentiation ability, with a strong inclination towards trophoblast lineage differentiation. These findings are significant and should be considered in future research endeavors. The objective of this study is to provide a comparative analysis of the pluripotency of pLCDM and pEPSC for a more comprehensive understanding. To comprehensively assess the pluripotency of pEPSC and pLCDM, low input RNA-seq was conducted on the cell lines. The transcriptome sequencing data of pLCDM (GSE197451) have been publicly published by this research team , while the transcriptome sequencing data of pEPSC established by our laboratory has not been uploaded, and this upload is the data upload of pEPSC.
