The coding region has been updated to extend the N-terminus to one that is more supported by available transcript and conservation data. It should be noted that an upstream ORF (uORF), which has a strong Kozak signal and encodes 2 aa, overlaps the updated start codon by 1 nt. This uORF may inhibit the use of the upstream start codon, and therefore allow the downstream start codon to be used via a reinitiation shunt mechanism. Alternatively, low level leaky scanning past the uORF start, or translation reinitiation due to the minimal overlap, may allow the upstream start codon to be used. There is no experimental evidence indicating which start codon is preferentially used in vivo. |
