SRX341237: Arbutus unedo nuclear Restriction-Site Associated DNA (RAD) fragments sequencing

Design: Arbutus unedo DNA was extracted from purified nuclei and digested with restriction enzymes Csp6I and TasI (Fermentas). Subsequently, end repair of fragments, ligation of adapters and nick repair were carried out using the Ion Plus Fragment Library Kit (Life Technologies), being the fragments purified after each reaction using silica column based kits. Finally, fragments were separated by agarose gel electrophoresis and size-selection (200 to 400bp) was made by gel slicing followed by purification using the GeneJET Gell Extraction Kit (Fermentas).
