Comparative analysis of RNA interference and pattern-triggered immunity induced by dsRNA in response to virus infection

(Submitter supplied) The main goal of this study was to compare the differential efficiency of the antiviral activity triggered by externally delivered virus-specific and nonspecific dsRNAs in plants. RNAi mediated by virus-specific dsRNA is the main antiviral defense mechanism in plants, but nonspecific dsRNA-triggered responses have been documented to play a role in antiviral defense. Using PVX as a model, we have determined that co-inoculation with either virus-specific or nonspecific dsRNA reduced virus accumulation in both inoculated and systemic leaves although at different extent. more...

Transcriptome analysis reveals the key role of overdominant expression of photosynthetic and respiration-related genes in the formation of tobacco(Nicotiana tabacum L.) biomass heterosis

(Submitter supplied) Heterosis, a phenomenon where hybrids exhibit superior performance in various traits such as yield, quality, and resistance compared to their parents, has long been a focus of agricultural research. Although tobacco biomass significantly affects tobacco yield and economic profitability, the underlying reasons for the dominance of tobacco biomass hybrids and their formation remain unclear. To address this, we selected 5 tobacco varieties (lines) with large differences in tobacco biomass as the parents and formulated F1 hybrids to analyze the expression of tobacco biomass heterosis. more...

Transcriptome analysis reveals the mechanism of action of tobacco NtAN3 in regulating leaf size formation

(Submitter supplied) Tobacco (Nicotiana tabacum L.) is an important cash crop, and the size of its leaves significantly influences both yield and quality. However, the upper part of tobacco leaves, due to its dense tissue structure, often faces issues such as narrow and thick leaves during the production of roasted cigarettes. These problems have a severe impact on the yield and quality of the upper leaf. Although the mechanism of leaf size regulation in Arabidopsis thaliana has been extensively studied, it remains unclear for tobacco. more...

Dual-transcriptomic analysis of Nicotiana benthamiana and two compatible Pseudomonas strains

(Submitter supplied) We sought to compare and contrast plant host and bacterial transcriptional changes during compatible infections that cause disease (albeit within different symptoms). We investigated the infection by the two Pseudomonas syringae sensu lato strains P. syringae pv. syringae B728a (Psy) and P. amygdali pv. tabaci 11528 (Pta) of Nicotiana benthamiana at an early time point post inoculation to understand how a plant host responds to two related bacteria with different infection strategies. more...

The transcriptional landscape of Nicotiana benthamiana and Verticillium dahliae interaction

(Submitter supplied) Verticillium dahliae is a soil-borne fungus with a broad host range, including the model plants Nicotiana benthamiana and Arabidopsis thaliana. The plant immunity can be activated by Vd-derived patterns while V. dahliae secreted effectors. We report here the RNAseq analyses of samples from N. benthamiana infected by V. dahliae strains 592 and 171 at different time points. The data showed dynamic expression patterns of genes not only from plant defense signaling but also plant developmental signaling.

Changes in Physiology of Burley Tobacco Plants Genetically Engineered to Express a Functional NtEGY2 Gene

(Submitter supplied) Nitrogen physiology is important in tobacco because of its role in generation of leaf yield and accumulation of nitrogen-containing alkaloids and that can react with nitrosating agents in the formation of carcinogenic tobacco specific nitrosamines. Cultivars of the burley tobacco market class are homozygous for deleterious alleles at the duplicate Yb1 and Yb2 loci which have previously been associated with decreased nitrogen use and utilization efficiency,; increased leaf nitrate, total nitrogen, and alkaloid levels,; and reduced yields. more...

Transcriptome analysis of different tissues in different materials

(Submitter supplied) We used two hybrid combinations and their 4 parents as materials. One week after topping the tobacco plant, samples were taken, including three tissues: root, stalk, and leaf. The RNA of these samples was extracted, and transcriptome sequencing analysis was carried out to explore the transcriptional profile differences between hybrids and their parents, and to analyze the gene differential expression between different tissues.

A woody plant degrading pathogen effector targets host glucose signaling protein VvRHIP1 to promote virulence

(Submitter supplied) Lasiodiplodia theobromae is one of the causal agents of Grapevine trunk diseases, which becomes a tremendous threat to the grapevine production worldwide. Plant pathogens secrete diverse effectors to suppress host immune responses or regulate the host metabolism to promote diseases. Our results suggest that L. theobromae LtCre1 targets host VvRHIP1 to manipulate the sugar signaling pathway, through disrupting the association of VvRHIP1 and VvRGS1 complex.

Suppression of pyrrolidine ring biosynthesis and its effects on gene expression and subsequent accumulation of anatabine in leaves of Tobacco (N. tabacum L.)

(Submitter supplied) Background: Anatabine, although being one of four major tobacco alkaloids, is never accumulated in high quantity in any of the naturally occurring species from the Nicotiana genus. Previous studies therefore focused on transgenic approaches to synthetize anatabine, most notably by generating transgenic lines with suppressed putrescine methyltransferase (PMT) activity. This led to promising results, but the global gene expression of plants with such distinct metabolism has not been analyzed. more...

Characterization of MdMYB68, a suberin master regulator in russeted apples.

(Submitter supplied) Apple russeting is mainly due to the accumulation of suberin in the cell wall in response of defects and damages in the cuticle layer. Over the last decades, massive efforts have been done to better understand the complex interplay between pathways involved in the suberization process in model plants. However, the regulation mechanisms which orchestrate this complex process are still under investigation. more...
