The p.I278K variant (also known as c.833T>A), located in coding exon 4 of the MSH6 gene, results from a T to A substitution at nucleotide position 833. The isoleucine at codon 278 is replaced by lysine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. The variant has been reported in ClinVar (Variation ID: 230128). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive.
